Protein S Activity, Plasma (HM0018)

Description

Protein S Activity, Plasma is a specialized coagulation test used to measure the functional activity of Protein S, a natural vitamin K–dependent anticoagulant protein that works as a cofactor for activated protein C to help prevent excessive blood clotting. This test is primarily used in the evaluation of inherited or acquired Protein S deficiency, unexplained venous thromboembolism (VTE), recurrent deep vein thrombosis (DVT), pulmonary embolism (PE), recurrent pregnancy loss, thrombophilia workup, and clotting disorders. Protein S activity testing helps identify reduced anticoagulant function and is often interpreted together with Protein S Free Antigen and Protein S Total Antigen for complete characterization of deficiency types. This test is important in thrombophilia assessment and anticoagulation-related diagnostic evaluation. Labcorp lists it as a test used for confirmation and characterization of congenital Protein S deficiency.

Pre-Test Instructions:

  • No fasting required
    Collect blood in light blue-top / blue-top tube (3.2% sodium citrate)
    Sample should be processed as platelet-poor plasma (double centrifugation recommended)
    Freeze plasma immediately after processing and transport frozen
    Avoid warfarin (Coumadin) for 2 weeks before testing if clinically appropriate, because it can lower Protein S levels/activity
    Avoid heparin for 2 days before testing if clinically appropriate; do not collect from a heparinized line/catheter
    If hematocrit >55%, the citrate volume in the collection tube may need adjustment
    Grossly hemolyzed, clotted, thawed in transit, or improperly labeled samples may be rejected
    Heparin or lupus anticoagulants may cause overestimation of Protein S activity, while APC resistance / Factor V Leiden may falsely lower measured Protein S activity
    Protein S activity is often interpreted together with Protein S Free Antigen and Protein S Total Antigen for complete thrombophilia evaluation and deficiency typing
