NK Panel (CD3, CD16, CD56, CD45) (HM0073)

Description

NK Panel (CD3, CD16, CD56, CD45) is a specialized flow cytometry blood test used to evaluate natural killer (NK) cells and lymphocyte subsets in peripheral blood. This immunophenotyping panel measures the expression of CD3, CD16, CD56, and CD45 markers to identify and quantify NK cells (typically CD3−/CD16+/CD56+), assess total lymphocyte populations, and support the evaluation of immune status, recurrent infections, immune deficiency disorders, autoimmune conditions, chronic viral infections, hematologic abnormalities, and immune monitoring. CD45 is used as a pan-leukocyte marker for gating lymphocytes, CD3 identifies T lymphocytes, and CD16/CD56 help characterize NK-cell populations. This test is commonly requested in clinical immunology, hematology, transplant workup, and immune function assessment. It provides both percentage and absolute counts of NK cells and related lymphocyte subsets, helping clinicians assess immune competence and monitor immune-related therapies. Flow cytometry is the standard methodology for NK-cell subset testing in major reference labs.C

Pre-Test Instructions:

  • Collect whole blood in EDTA (lavender-top) tube
    Some labs may additionally require ACD-A / ACD-B (yellow-top) tube depending on the exact panel and reference laboratory protocol
    Fill tube adequately to preserve cell viability for flow cytometry
    Invert tube 8–10 times immediately after collection to mix anticoagulant properly
    Do not refrigerate or freeze specimen; maintain at room temperature only
    Sample should ideally reach the testing laboratory within 24–48 hours of collection for accurate cellular analysis; some labs require arrival within 48 hours, while others prefer shipping within 24 hours depending on assay type
    Avoid clotted, hemolyzed, contaminated, or delayed specimens as these may be rejected or give unreliable results
    Mention date and time of collection on the tube/requisition when sending to a reference lab
    If the patient has severe lymphopenia, some advanced NK subset assays may be canceled or need alternate lymphocyte subset testing because a minimum CD45 lymphocyte count is required
