Anti-Sperm Antibody (ASAB) (IO0036)

Description

Anti-Sperm Antibody (ASAB) is a specialized fertility-related immunological test used to detect antibodies directed against sperm cells in serum or semen, which may interfere with sperm motility, sperm transport, sperm-cervical mucus interaction, fertilization, and overall reproductive potential. This test is commonly used in the evaluation of male infertility, immunological infertility, unexplained infertility, low sperm motility (asthenozoospermia), sperm agglutination, post-vasectomy or post-vasectomy reversal fertility issues, and in selected female infertility workups where antisperm antibodies may also be suspected. Antisperm antibodies may form after testicular trauma, infection, surgery, obstruction, vasectomy, or disruption of the blood-testis barrier, and they may reduce the ability of sperm to move normally or fertilize the egg. Clinical review literature describes two major direct semen-based methods for detecting sperm-bound antibodies: the Mixed Antiglobulin Reaction (MAR) test and the Immunobead test (IB test), typically assessing IgG and/or IgA antibodies. Serum-based ASAB testing is commonly performed by ELISA / EIA, while semen-based testing is often reported as percentage of motile sperm bound by antibodies or positive/negative depending on method. This test is usually used as part of a broader male infertility workup alongside semen analysis, sperm motility evaluation, and reproductive specialist consultation.
